Question about gain

when using a dmm to set your gain properly on your amp, how many volts are you supposed to look for? I watched the ed video and they used ~40, but why 40? What would I use for a basis? Thanks. Also what else do I have to do, I think someone said it in an earlier thread, but turn volume 3/4 of the way, turn off some stuff on hu, just need a refresher

Turn sub level all the way up. Turn off EQ and set any other HU settings to 0 or off. Turn off bass boost.
