Does Positive and Negative matter on the speakers?

From my 4ch amp if I have the positive and negative mixed up for the door speakers, will this create problems?

it may. say if you have yer left speaker wired correctly and yer right speaker wired backwards (the speakers would be wired "180 degrees out of phase") it may cause cancellation issues. it's always a good idea to wire them correctely. but in the end, if you wire it backwards, nothing will break or blow or sound bad or anything.

it makes a huge difference. as stated the speakers will fire out of phase. meaning one will push while the other pulls. you want to control the performance of the drivers so that they reproduce the sounds as accurate to the source as possible. so, changing the polarity is often done intentionally to achieve the disired effect. it depends on your needs. it can also make your system sound poorly.

but yes, it does most certainly "matter".
