What is the main difference between the JBL BTX 250 or another tube woofer and a regular sub in a box?

Difference as in SQ? Besides the physical difference, not much performance wise. Most people with limited space buy tube subs.
For best SQ, a sub in a proper box will sound much better.

It sounds like a decent product. I'm not really into tube subs cause I didn't find one that fit my quality standard.

You'll need 12v power supply (around 15A-20A)since it has built in amp. That's about it.

Well you will need rca cable to connect it to the tube sub but that's given.
You don't need a splitter unless you're planning on sharing the dvd player's output with another amp.

Converter as in power supply? You can get one from most electronic/hobby stores. eBay has it too. Look for any 12v power supply with 15A-20A. The cost will vary anywhere from $30 - $100.
You could build your own for probably less.
